Per Hackaday, researchers have demonstrated allele-specific antisense therapy in two boys with severe epilepsy and developmental delays caused by SCN2A mutations. This precision approach targets only the faulty gene copy, leaving the healthy one intact—a critical distinction that standard anti-seizure medications can't make. The results were striking: in the nine-year-old, twelve doses over two years eliminated the need for one medication while reducing seizures overall, and in the fourteen-year-old, eight doses over sixteen months dropped his daily seizures from roughly two to zero. Beyond seizure control, both boys showed measurable gains in language, motor skills, and behavior—developmental progress that opens the door to earlier, more targeted intervention in future cases.
